designed to help law enforcement combat technology-facilitated child
exploitation crimes. It will gather detailed national data on a
number of policy and practice relevant topics including: 1) new
developments and numerical trends in cases coming to law enforcement
attention; 2) what investigative strategies are associated with more
favorable outcomes; 3) what challenges and dilemmas confront
prosecutors; 4) what indicators and investigative procedures are
more likely to identify child pornography offenders who have also
committed crimes against or endangered children in their
environments; and 5) how investigators are managing and responding
to cases of sexual images produced and disseminated by youth. The
data will be gathered using a proven, successful methodology, a
2-stage survey of a national sample of law enforcement agencies that
will include interviews with investigators and prosecutors involved
in a representative sample of more than 1000 technology-facilitated
crimes. The research will be conducted by the team with the nation’s

NEW MEXICO Mycobacterium
tuberculosis latently persist in 2 billion people, representing a
major reservoir of tuberculosis (TB) due to reactivation, yet the
state of latent bacilli remains a mystery. In order to eradicate TB,
we need to understand latency and develop in vitro models for drug
screening. In this interdisciplinary project we have merged
microbiological tuberculosis research with nanotechnology and novel
materials. Recent publications demonstrate a unique phenomenon of
cell-directed assembly of hybrid lipid-inorganic nanostructures that
drive a unique non-replicative persistence, and protect bacteria and
drastically prolong viability. We show these same structures can be
formed using mycobacteria, and that they display the highly extended
viability characteristic of latency. We hypothesize that the
persistence of latent TB, and its resistance to chemotherapeutic
challenge, can be modeled by incorporation of bacilli into
self-assembled lipid-inorganic nanostructures. This model combines
the ability for in vitro study and growth condition modulation, with
the potential for in vivo implantation and infection. We will
generate a new in vitro model for high throughput screening for
targets of, and drugs that can eliminate latent TB, to complement
the existing complex in vivo models and al- low this field to move
forward. Specific Aim 1 Define the conditions for optimal
lipid-inorganic nanostructure-encasement of mycobacteria and its
effects upon mycobacterial resistance to drugs. Specific Aim 2
Determine the changes in mycobacterial metabolism occurring upon
encasement in lipid- inorganic nanostructures that result from
non-replicative persistence, and drive in vitro survival and
latency. Specific Aim 3 Demonstrate that lipid-inorganic
nanostructure-encased mycobacteria represent valid models of latency
by showing their infectivity in mice, even after extended periods of
time after assembly. PUBLIC HEALTH RELEVANCE: Public Health
Relevance We need to understand the way that tuberculosis is able to
exist in a latent state because it is present in 2 billion people
worldwide and reactivates into an active form in a significant
percentage of cases. Clearing this 'reservoir' will be an essential
component of an eventual eradication strategy. Although there are
animal models of latency, there is no good model of in vitro
latency, yet we need to have such an in vitro model so that we can
understand how to attack it, and also to screen drugs with. We have
developed a unique nanostructured model that mimics the extreme
non-replicative persistence of latency, and want to test how

UNIVERSITY Chlamydia trachomatis, the
most common bacterial sexually transmitted disease (STD) agent
worldwide, often establishes asymptomatic genital infections which
result in serious reproductive complications. In culture,
environmental stressors can induce chlamydiae to enter a viable but
non-infectious state termed "persistence". Persistent
chlamydiae: i) can re-enter the normal developmental cycle when the
environmental stressor is removed and ii) are refractory to killing
by anti-chlamydial drugs. Persistent organisms may also contribute
to the long-term inflammation and scarring characteristic of
chlamydial diseases by chronically releasing pro-inflammatory
effectors and/or by serving as a reservoir for continued cycles of
productive infection. Though there is indirect evidence that the
persistent state occurs in vivo, direct proof for any role of
persistent organisms in chlamydial pathogenesis has remained
elusive. The goal of this proposal is to develop an experimentally
tractable animal model of persistent chlamydial infection so that
the role of persistent organisms in chlamydial pathogenesis can be
assessed. Unfortunately, most of the currently characterized
persistence inducers (nutrient starvation, IFN-3-exposure, etc.)
would be impossible to reproducibly employ in an animal model
system. Recent studies in our laboratory indicate that exposure to
amoxicillin, the most commonly prescribed 2-lactam in the US, as
well as clavulonate, induces C. trachomatis persistence in culture.
Additional investigations using C. muridarum-infected mice and C.
psittaci- infected cats suggest that amoxicillin/clavulonate
(Amox/Clav)-exposed animals develop persistent infection. Amox/Clav
is an ideal persistence inducer for use in an animal model as it: i)
is relatively non-toxic, ii) acts directly on the developing
chlamydiae and iii) can be easily added to or subtracted from the
system. Therefore, we will establish an in vivo murine model of C.
muridarum persistence by testing the following two hypotheses in
two, related, but independent, Specific Aims. Hypothesis 1:
Amox/Clav-exposed, persistent chlamydiae can induce alterations in
murine host cell gene expression (Aim1). Hypothesis 2:
Amox/Clav-exposure can induce persistent chlamydial infection in an
in vivo murine model system (Aim 2). Completion of these Aims will
not only test both hypotheses but will also provide a necessary
foundation for future studies regarding the role of persistent
chlamydiae in chronic disease induction. Chlamydia trachomatis is
the most commonly reported bacterial STD agent in the US (4,000,000
new cases/year) and worldwide (90 million new cases/year). In women,
C. trachomatis genital infection often results in salpingitis,
sterility or atopic pregnancy. Genital chlamydial infections cost an
estimated $10 billion in the US in 2002. This proposal will develop
an experimentally tractable animal model of chlamydial persistence,
which will ultimately allow us to determine how persistent infection
impacts pathogenesis, transmission and antimicrobial therapy for
these infections. PUBLIC HEALTH RELEVANCE: Chlamydia trachomatis is

a common cause of sterility and potentially fatal atopic pregnancy -

Alarmingly, despite the development of excellent screening tests and
effective anti-chlamydial drugs, the incidence of chlamydial genital
infections in the US continues to rise. This study will determine
whether or not chlamydiae can establish persistent infections in the
genital tract, which has important implications for development of

BE NON-TOXIC AND NON-FLAMMABLE, BUT ADDITIVES WILL BE INCLUDED TO
PRECLUDE DETERIORATION OF WITHER THE PCM CONTAINER OR THE DIAPHRAGM
MATERIAL. THE USE OF A WATER-BASED PCM GIVES THE HIGHEST HEAT
CAPACITY FOR THE MASS. THIS IS HIGHLY LIMITED DUE TO THE NEEDS FOR
PORTABILITY AS REQUIRED FOR AN EXTRA-VEHICULAR ACTIVITY (EVA). THE
TOTAL HEAT CAPACITY OF AN OPERATIONAL UNIT WOULD BE FOR 4 HOUR
DURATION EVA USE. THROUGH A LOGICAL PROGRESSION OF TASKS INCLUDING
CONCEPT OF OPERATION FORMULATION, REQUIREMENTS FORMULATION, CONCEPT
DESIGN REVIEWS AND DETAIL DESIGN REVIEWS THAT INCLUDE DESIGN AND
THERMAL ANALYSIS USING THERMAL DESKTOPTM MODELS, THIS EFFORT CAN
PROGRESS FROM THE TRL 3 ACHIEVED IN PHASE I TO TRL 4-5. THE TEAM
WILL CONTINUE DEVELOPMENT BY DESIGNING A VARIABLE CONDUCTANCE
INTERFACE (VCI) FOR PROTECTING WATER IN THE LIQUID COOLING GARMENT
(LCG) FROM FREEZING DUE TO THE TEMPERATURE OF THE HEAT SINK USED BY
THE PCM. THE TEAM WILL ALSO DEVELOP SYSTEM IMPROVEMENTS IDENTIFIED
DURING PHASE I TESTING. THE PCM WILL BE TESTED TO CONFIRM HEAT
INPUT/TEMPERATURE PERFORMANCE AND CYCLING CAPABILITY. THE TEST BED
WILL ALLOW FOR ACCURATE HEAT INPUT KNOWLEDGE, TEMPERATURE MONITORING
AND CYCLING CAPABILITY. THE RESULTS WILL BE COMPARED TO THE THERMAL
MODEL TO ENSURE ACCURATE PREDICTION CAPABILITY FOR THE NEXT PHASE
UNIT AND SYSTEM IMPLEMENTATION. THE DESIGN DESCRIPTION AND TEST
