Coastal and Marine Habitats to Benefit: The target shoreline of Piscataway Park fronts a 10-acre tidal marsh. The Potomac River area has prime sportfishing and is under severe threat of habitat loss. The shoreline and marsh are rich in submerged and emergent aquatic vegetation (SAV and EAV), which provide nursery habitat for anadromous fish, forage fish, mussels, birds, and butterflies. The living shoreline design will enhance the nursery area to encourage greated aquatic species diversity and numbers, as well as SAV colonization. Threatened and endangered species found in the Potomac River nearby include short nose sturgeon, bald eagles, yellow lampmussel, and the Baltimore checkerspot butterfly. Rare waterfowl includes snow goose, brandt, blue-wing teal, redhead duck, greated scaup, long-tail duck, and common goldeneye. Pumpkin ask ls located in the northern and southern sections of the target shoreline and is on Maryland's list of current and historical rare, threatened, and endangered species. Project Scope: A vegetated "living shoreline," using both structural and nonstructural techniques, will be constructed on 2,800 feet of Potomac River shoreline. The northern coast of the target shoreline will include construction of stone spurs and stone wills, with sand sill and wetland plantings along approximately 1,000 feet of potomac River shoreline. Porject Outuput/Outcomes: 1) 2,800 linear feet of Potomac River shoreline will be protected and restored; 2) the opening to a 10-acre wetland will be protected; 3) important fish and wildlife shoreline habitat will be restored and preserved; 4) threatened Native American archeological resources at the Accokeek Creet Site, shich is located at the confluence of Piscataway Creek and the Potomac River, will be protected; 5) the viewshed from Mount Vernon, which is located directly across the Potomac River, will be protected; 6) water quality will improve locally; 7) AFF non-profit staff jobs will be preserved; 8) construction, nursery, and professional consultant jobs will be supported.

The Athapascan or, as speakers prefer, Dene languages constitute the largest and most geographically widespread language family of native North America. Five separate sub-groupings have been identified: Alaskan, Northern Interior (BC/Yukon), Mackenzie Basin, and the non-continuous Pacific Coastal and Apachean groups. All of the Dene speech communities are endangered through the enormous economic and social pressures that weight on tribal people throughout North America. Fluency is rapidly disappearing. However, the Dene communities are aware of their fragility and many are actively working with linguists to document their languages. This proposal seeks funding for the development of a online interactive speech atlas of the Dene speaking communities in the Mackenzie Basin by team of tribal and academic linguists and educators. The Speech Atlas will have clickable links to the Dene speaking communities listed on the map. The links to information about the properties of speech that underlie structure of the sound system in that community. These will include a list of the inventory of speech sounds for that community, written in two orthographies, the International Phonetic Alphabet and in the orthography developed for use in that community. Sound files of words exemplifying the consonant and vowel speech sounds will be given. Information on tone and prosody will be collected and presented. The Atlas will make available baseline information on intonation patterns in questions and statements, demonstrating basic discourse patterns. A desirable feature of this approach is the ability to localize Dene language data to a specific community, reflecting the way the Dene identify themselves (linguistic identity is strongly community-based) and highlighting points of variation and stability across the languages of the characteristically small and often isolated Dene communities in this area. The Dene Speech Atlas project will foster integration of research and education through the teams' ongoing associations to CILLDI (Canadian Indigenous Language and Literacy Development Institute), held at University of Alberta, Edmonton every summer. The Institute is devoted to training speakers of indigenous languages in linguistic analysis and language documentation, drawing speakers from indigenous communities all over northern Canada.

Combining optical and near-infrared data, the team will estimate how the rate of star formation and the stellar masses of galaxies changed over this period, which covers almost half of cosmic history. In the twenty-first century, cervical cancer prevention programs represent the expenditure of significant public health dollars. In anticipation of changes in cervical cancer prevention strategies such as first- and second- generation human papillomavirus (HPV) vaccines and in the longer-term extended Pap test screening intervals and novel HPV test applications, the New Mexico Notifiable Diseases and Conditions list incorporated mandatory state-wide reporting of all Papanicolaou (Pap) tests, cervical pathology results and HPV tests. Currently, New Mexico is the only state with the capacity to fully monitor cervical cancer screening and prevention using a woman-based information system. This research program embodies a sustainable US model cervical cancer screening registry. Baseline population-based screening behaviors and disease incidence will be characterized to allow future assessment of changes in state-wide cervical cancer screening programs. The two main peaks of HPV-related population events including the peak of HPV infection occurring in women between the ages of 18-22 and the peak of HPV-related cervical pre-cancers occurring in women between the ages of 25 and 30 will also be characterized. Broad spectrum HPV genotype- specific prevalence will be determined in a population-based sample of residual liquid cytology specimens (n=60,000) prior to mass HPV vaccine implementation. These data will allow an assessment of the longitudinal HPV genotype-specific risk for high-grade cervical disease (eCIN2/3 or AIS) outcomes over extended periods of our state-wide surveillance program. Vaccine and non-vaccine HPV genotypes will also be characterized in a population-based sample of tissues from women diagnosed with low-(CIN1; n=5,500) and high-grade (eCIN2/3 or AIS; n=4,500) cervical intraepithelial neoplasia prior to and overlapping with initial HPV vaccine introduction. A surveillance registry with the capacity to monitor the overall and type-specific HPV incidence of cervical precancers as well as screening behaviors will enable longitudinal assessment of actual effectiveness of interventions decades before any potential impact on cervical cancer incidence will be realized. These research data are critical to determining the public health significance and cost effectiveness of interventions impacting HPV-related cervical disease incidence. Furthermore, the surveillance and research data will enable a determination of any unforeseen outcomes such as replacement of vaccine HPV types by non-vaccine HPV types or changes in screening behaviors among vaccinated and unvaccinated women. Without conducting the population-based screening surveillance proposed by our program, these determinations are simply not possible. In the background of highly successful US cervical cancer screening programs, investment in this research program represents due diligence as we move forward with promising new primary and secondary interventions for cervical cancer for which much remains unknown. PUBLIC HEALTH RELEVANCE: In the interest of the health of all women, our research program will conduct longitudinal population- based surveillance in New Mexico to assess the overall benefits and risks associated with HPV vaccines and other cervical cancer prevention measures. New Mexico offers a rich opportunity to conduct this research in diverse multi-cultural populations that suffer many health disparities including those attributable to high levels of poverty and rural living. In the background of the already highly successful US cervical cancer screening programs with an estimated cost of over $5 billion annually, investment in this research program represents due diligence.

REGENTS OF THE UNIVERSITY OF MINNESOTA


Although fusion of sperm and egg to form a new organism was first observed microscopically over 160 years ago, the proteins on the surface of the sperm and egg that facilitate this process are still largely unknown. Over the last decade, the small (~ 1mm) nematode worm C. elegans has been used to discover seven sperm proteins and six egg proteins that are required for fertilization to take place. The focus of this proposal is SPE-42, a protein that, when mutated, results in complete sterility of the affected worms. Preliminary data, including computer models, suggest that SPE-42 is expressed on the sperm surface, meaning that SPE-42 is potentially available to interact with proteins present on the egg surface. The experiments to be carried out will test this hypothesis by asking (1) where in sperm is SPE-42 located and (2) which parts of the protein are important for its function. A combination of molecular genetic techniques and computer modeling of the SPE-42 protein structure will be used to complete the proposed work. In addition, a collection of new fertilization-defective mutants will be evaluated to find other proteins that are involved in sperm ? egg interactions during fertilization. The knowledge gained from these experiments will impact both the fields of infertility treatment and non-hormonal contraceptive development. Americans spend nearly $1.4 billion per year on infertility treatment, and most of the bill is not covered by medical insurance. Because proteins similar to SPE-42 are found in nearly all multi-cellular organisms including humans, what we learn about how SPE-42 works will inform our understanding of animal fertilization in general. This project will provide hands-on training to at least two graduate students and several undergraduates.

The goal of this project is an enhanced understanding of the mechanisms by which plants acquire and then maintain the correct levels of iron and other metal nutrients. Currently, little is known about molecules involved in the movement of metals on a large scale, for example, from roots to leaves, or from leaves and roots into seeds. Knowledge of the mechanisms involved in plant iron movement is lacking, presenting a major obstacle to devising approaches for biofortification of staple foods. Biofortification refers to the modification of crops to accumulate additional iron in edible parts; it is widely regarded as a sustainable means of improving the iron nutrition of the 2-3 billion people worldwide whose inadequate diet causes iron deficiency anemia. Improving our understanding of plant iron homeostatic mechanisms is also critical if we wish to improve growth of crops in marginal soils, where iron deficiency frequently limits crop growth. The investigators' work on the Yellow Stripe-Like (YSL) family of proteins is focused on these large scale, whole plant processes: YSL transporters are required for normal movement of iron, zinc, and copper in both vegetative and reproductive tissues. By analyzing mutant plants that lack one or more YSL genes, the investigators will elucidate the key YSL genes that mediate large scale movements of metals in plants. By analyzing global gene expression in mutant plants, the investigators will identify novel genes and gene networks that are involved in plant Fe homeostasis. The investigators will also develop more reliable and straightforward assays that characterize the transport activities of the YSL transporters. This project will provide numerous (at least 60 over the three year duration of the project) meaningful opportunities for undergraduate participation in this research program, and in addition, will educate and train a PhD student and two postdoctoral students.

CASE WESTERN RESERVE UNIVERSITY


Dr Zehavi will analyze and model the clustering of galaxies in large surveys such as the Sloan Digital Sky Survey (SDSS), to probe galaxy formation and the development of cosmic large-scale structure. She and others have shown that on small scales, dim red galaxies are more strongly clustered than brighter ones. This could be explained if the dim galaxies are satellites of more luminous galaxies, sharing their massive dark halos. Dr Zehavi and her team will test this hypothesis by examining much larger samples from the SDSS, and comparing the clustering properties of the dim red galaxies to those predicted for satellite dark halos by gravitational N-body simulations. The SDSS data will also be used to examine the distribution of satellite galaxies, comparing the observed anisotropic distribution with predictions from current models in which the satellites form in filaments of the cosmic web. She will use other surveys of more distant galaxies to examine how galaxy clustering has changed over the past 10 billion years of cosmic history. To do this, Dr Zehavi will construct a Halo Occupation Distribution model, which gives a statistical relation between the luminosity and appearance of a galaxy at a given time and the mass of its dark halo. The growth of dark halos can be followed in cosmological N-body simulations, so this model will provide a link between present-day galaxies and their probable precursors, billions of years earlier. A postdoctoral scholar will be trained by participating in the research; undergraduate students will also be involved. Dr Zehavi will continue to give public-outreach talks at local venues. She will partner with a local museum and planetarium to develop exhibits, and mentor undergraduates working as museum interns.
